How estrogenic is PM in comparison to E2?

Bimodal action of miroestrol and deoxymiroestrol, phytoestrogens from Pueraria candollei var. mirifica, on hepatic CYP2B9 and CYP1A2 expressions and antilipid peroxidation in mice.

The aim of the present study is to investigate the impact of miroestrol and deoxymiroestrol (Fig. 1) on the regulation of hepatic CYP2B9 and CYP1A2 at both transcriptional and enzymatic levels. We hypothesized that because miroestrol and deoxymiroestrol have similar structures compared with estradiol and possess estrogenic-like activities, these 2 compounds should regulate mouse hepatic P450s in a similar manner as estradiol. Furthermore, the antilipid peroxidation activities of these 2 compounds were determined in the mouse brain to provide additional beneficial biologic activities.

Hi Denita,

Excellent post,

This is so important to understand and vital for breast growth, if this were more easily understood we'd be miles ahead in terms of how to utilize this knowledge.

Quote:This is the first report suggesting miroestrol and deoxymiroestrol as potential alternative medicines to estradiol because of their distinctive ability to regulate mouse hepatic P450 expression and their beneficial antioxidative activities.

You'll find more info about enzymes in the FAQ, one more thing, certain herbals can affect plasma concentrations, I'd talk more about them but it's still something that's kinda new for NBE. (Which btw I've tried already).

Lotus I think this needs to be in the Bio Male FAQ too no???

Yes, great idea EvaMarie!, Wink

We'd need the approval from the OP, Eloise what do you think?.

Its your thread on the female NBE FAQ is what I meant... It applies here for Bio males too right???

Im sure lots of us never make it over there....

Not to take away from the OP's thread of course....

Thanks Lotus. Smile

I am looking at a several studies on a mineral effects in decreasing SHBG and the subsequent increase of free Progesterone.

Also have a paper on a naturally induced response to increase HGH.

I will read up on the Nutrition Research Journal post when i get time.

If its ok with you can I run all finding past you before I post anything for a your opinion and a validity check?

Hi Denita,

That's awesome, SHBG is so poorly understood, I think explaining why we want reduce it and how it will benefit our BMI is a great idea just for starters. Also, explaining how it binds to sex hormones would be helpful.

Sweetheart you post whatever you'd like, and I'll enjoy reading and commentating on them. The only thing I would recommend (or request) is that any research you find on anything related to hormones is if you can add them into the new Endocrine section, but that's strictly up to you, I won't be offended the least. Wink

Everything else sounds promising!

1) In addition to miroestrol and deoxymiroestrol, Pueraria Mirifica also contains other chemicals that belong to isoflavones and Coumestan groups of phyto-estrogens, e.g.,
  • Genistein,
  • Daidzein,
  • Daidzin,
  • Genistin,
  • Coumestrol
  • Miroestrol
  • Deoxymiroestrol

2) Miroestrol (the bioactive component of Pueraria Mirifica) appears to be a quarter as potent as 17β-estradiol in inducing estrogenic like effects. It has been noted that Deoxymiroestrol is actually stronger than Mirosterol (as well as Isomiroestrol) with about 10-fold greater potency in an MCF-7 cell assay relative to Miroestrol. These authors suggested that Deoxymiroestrol should be considered the main bioactive estrogen, since it is plausible that passive oxidation by exposure to the air could have resulted in Miroestrol production from Deoxymirosterol. Relative to the Soy Isoflavones Genistein and Daidzein, the chromene compounds Miroestrol and Deoxymiroestrol are approximately 1,000-10,000 fold more potent on a weight basis (claim stated here, which compares the potencies derived from these two studies[10][12]) and in some studies directly assesing the claim it is found that the soy isoflavones do not contribute towards the majority of estrogenicity.

3)-The chromene compounds are potent and direct estrogen receptor agonists, and Deoxymiroesterol appears to be more potent than 17β-Estradiol itself (17β-Estradiol is the most relevant naturally occuring estrogen compound in the human body)

4) May have similar (in phenotype and potency) anti-androgenic and suppressive effects in the testes as estrogen itself:
-Isolated Miroestrol and Deoxymiroestrol incubated in testes of male mice appear to have similar suppressive effects on enzymes within the steroid synthesis metabolic pathway (17β-HSD1 and CYP19 mRNA, to a lesser degree 3β-HSD and CYP17) while inducing 17β-HSD2 mRNA; the direction of effects and magntiude of effect were similar when comparing the chromenes against estradiol.[61]

5) By modulating androgen receptor coactivators, daidzein may act as a phytoandrogen

(Oh yeah, we'll need to talk about this one!).
