Influence of virgin coconut oil-enriched diet on the transcriptional regulation of fatty acid synthesis and oxidation in rats - a comparative study.

Author information

Abstract
The present study was carried out to evaluate the effects of virgin coconut oil (VCO) compared with copra oil, olive oil and sunflower-seed oil on the synthesis and oxidation of fatty acids and the molecular regulation of fatty acid metabolism in normal rats. Male Sprague-Dawley rats were fed the test oils at 8 % for 45 d along with a synthetic diet. Dietary supplementation of VCO decreased tissue lipid levels and reduced the activity of the enzymes involved in lipogenesis, namely acyl CoA carboxylase and fatty acid synthase (FAS) (P< 0·05). Moreover, VCO significantly (P< 0·05) reduced the de novo synthesis of fatty acids by down-regulating the mRNA expression of FAS and its transcription factor, sterol regulatory element-binding protein-1c, compared with the other oils. VCO significantly (P< 0·05) increased the mitochondrial and peroxisomal β-oxidation of fatty acids, which was evident from the increased activities of carnitine palmitoyl transferase I, acyl CoA oxidase and the enzymes involved in mitochondrial β-oxidation; this was accomplished by up-regulating the mRNA expression of PPARα and its target genes involved in fatty acid oxidation. In conclusion, the present results confirmed that supplementation of VCO has beneficial effects on lipid parameters by reducing lipogenesis and enhancing the rate of fatty acid catabolism; this effect was mediated at least in part via PPARα-dependent pathways. Thus, dietary VCO reduces the risk for CHD by beneficially modulating the synthesis and degradation of fatty acids.

Why Coconut Oil Is So Good For Pregnant & Nursing Mothers
http://www.modernmom.com/c3443748-3b35-1...4a41e.html

Coconut oil contains large amounts of lauric acid, a powerful anti-microbial fatty acid that protects the immune system of the fetus and newborn. Pregnant and nursing mothers should eat coconut oil to increase the quality of the womb environment and breast milk” - Dr. Claudia Pillow

healthy diet contains mixtures of saturated, monounsaturated and polyunsaturated fats. The unique composition of human breast milk contains about 45 to 50 % saturated fat, about 35% unsaturated and 15-20%polyunsaturated. Lauric acid and capric acid comprise about 20% of total saturated fatty acids found in breast milk. Lauric and capric acid have potent antiviral, antibacterial, and parasiticidal (kills parasites) properties that support the immune system. These fatty acids offer the nursing infant protection from illnesses, viruses such as herpes and HIV, protozoa such as giardia lamblia, and bacteria such as chlamydia and heliocobater.

Coconut oil is high in saturated fat (but not cholesterol since it is from a plant) containing about 50% lauric acid. Other components of coconut oil include capric acid, caprylic acid, tocopherols and tocotrienols. (Vitamin E lipids that act as potent anti-oxidants that can help maintain healthy cell structure and function). A study published in the American Journal of Clinical Nutrition has shown that lactating mothers who eat coconut oil and other coconut products, have significantly increased levels of lauric acid and capric acid in their breast milk, creating milk rich in health promoting nutrients. Coconut oil is easy for an infant’s immature digestive system to absorb and utilize. It also provides energy in the form of medium chain fatty acids to help the baby grow and develop properly.

IDS 89 and its subfractions on 5 alpha-reductase activity in human benign prostatic hyperplasia.

Abstract
Extract from fruit of Sabal serrulata are used in the treatment of human benign prostate hyperplasia (BPH). Therefore, it is of interest whether this phytopharmacon has any influence on the androgen metabolism in the human prostate. It was found that the extract IDS 89 of Sabal serrulata inhibited dose dependently 5 alpha-reductase activity in the epithelium and stroma of human BPH, the mean inhibition being 29% and 45%, respectively. This inhibitory effect is mainly due to the saponifiable subfraction of IDS 89 showing a mean 5 alpha-reductase inhibition of 39% and 38% in epithelium and stroma, respectively. The inhibition was dose dependent and noncompetitive. At a testosterone concentration of 580 nM as substrate for 5 alpha-reductase, the main fatty acids of the extract IDS 89 gave rise to a percentual enzyme inhibition in the epithelium and stroma as follows: 51% and 42% (lauric acid), 5% and 0% (oleic acid), 43% and 34% (myristic acid), 2% and 0% (palmitic acid), respectively. The inhibitory effect of lauric acid was noncompetitive and dose dependent up to a concentration of 0.2 nM, the maximal inhibition in the epithelium and stroma being 52% and 45%, respectively. The nonsaponifiable subfraction, consisting mainly of phytosterols, showed a mean inhibition of 5 alpha-reductase in the epithelium and stroma of 15% and 10%, respectively. Finally, the hydrophilic subfraction, containing carbohydrates, amino acids, and polysaccharides, showed no inhibitory effect. The present in vitro studies suggest that the Sabal serrulata extract IDS 89 has an inhibitory effect on 5 alpha-reductase in the epithelium and stroma of human BPH. This inhibition is mainly due to the fatty acids of the saponifiable subfraction.

The enzyme encoded by this gene is a multifunctional protein. Its main function is to catalyze the synthesis of palmitate from acetyl-CoA and malonyl-CoA, in the presence of NADPH, into long-chain saturated fatty acids. In some cancer cell lines, this protein has been found to be fused with estrogen receptor-alpha (ER-alpha), in which the N-terminus of FAS is fused in-frame with the C-terminus of ER-alpha. More scientific proof that fatty acids helps with the binding process of estrogen receptor alpha (ER-a, the growth receptor). Specifically---palmitate (a solid unsaturated fat). I hope this is making sense for everybody. In other words- omega 3's and 6's needs to be in an NBE plan, the health benefits alone are worth looking into, another benefit is its ability to block DHT (e.g. Evening primrose oil, EPO).

Inhibition of steroid 5 alpha-reductase by specific aliphatic unsaturated fatty acids.

Human or rat microsomal 5 alpha-reductase activity, as measured by enzymic conversion of testosterone into 5 alpha-dihydrotestosterone or by binding of a competitive inhibitor, [3H]17 beta-NN-diethulcarbamoyl-4-methyl-4-aza-5 alpha-androstan-3-one ([3H]4-MA) to the reductase, is inhibited by low concentrations (less than 10 microM) of certain polyunsaturated fatty acids. The relative inhibitory potencies of unsaturated fatty acids are, in decreasing order: gamma-linolenic acid greater than cis-4,7,10,13,16,19-docosahexaenoic acid = cis-6,9,12,15-octatetraenoic acid = arachidonic acid = alpha-linolenic acid greater than linoleic acid greater than palmitoleic acid greater than oleic acid greater than myristoleic acid. Other unsaturated fatty acids such as undecylenic acid, erucic acid and nervonic acid, are inactive. The methyl esters and alcohol analogues of these compounds, glycerols, phospholipids, saturated fatty acids, retinoids and carotenes were inactive even at 0.2 mM. The results of the binding assay and the enzymic assay correlated well except for elaidic acid and linolelaidic acid, the trans isomers of oleic acid and linoleic acid respectively, which were much less active than their cis isomers in the binding assay but were as potent in the enzymic assay. gamma-Linolenic acid had no effect on the activities of two other rat liver microsomal enzymes: NADH:menadione reductase and glucuronosyl transferase. gamma-Linolenic acid, the most potent inhibitor tested, decreased the Vmax. and increased Km values of substrates, NADPH and testosterone, and promoted dissociation of [3H]4-MA from the microsomal reductase. gamma-Linolenic acid, but not the corresponding saturated fatty acid (stearic acid), inhibited the 5 alpha-reductase activity, but not the 17 beta-dehydrogenase activity, of human prostate cancer cells in culture. These results suggest that unsaturated fatty acids may play an important role in regulating androgen action in target cells.

Anti-androgenic activity of fatty acids.

In this study, we show that 5alpha-reductase derived from rat fresh liver was inhibited by certain aliphatic free fatty acids. The influences of chain length, unsaturation, oxidation, and esterification on the potency to inhibit 5alpha-reductase activity were studied. Among the fatty acids we tested, inhibitory saturated fatty acids had C12-C16 chains, and the presence of a C==C bond enhanced the inhibitory activity. Esterification and hydroxy compounds were totally inactive. Finally, we tested the prostate cancer cell proliferation effect of free fatty acids. In keeping with the results of the 5alpha-reductase assay, saturated fatty acids with a C12 chain (lauric acid) and unsaturated fatty acids (oleic acid and alpha-linolenic acid) showed a proliferation inhibitory effect on lymph-node carcinoma of the prostate (LNCaP) cells. At the same time, the testosterone-induced prostate-specific antigen (PSA) mRNA expression was down-regulated. These results suggested that fatty acids with 5alpha-reductase inhibitory activity block the conversion of testosterone to 5alpha-dihydrotestosterone (DHT) and then inhibit the proliferation of prostate cancer cells.

Adding another option for CT (cold thermogenesis) NBE, actually it's a blend of hot/cold therapy. Call it SPA BOOBs, this one you do in the shower as follows:

1-2 cups of crushed ice (per breast) for cupping method
1-2 ice packs.

Alternate hot water from the shower heating up the breasts, (1-2 minutes), face back to the shower and apply cup full of shredded ice, start in a clockwise position using the cup (1-2 minutes), alternate back to hot water heating breasts after completing the clock method, (15-20 seconds per 15 min. clock position), massage massge massage while completing this step, lol. Ultimately, a thermogenesis is taking place. Bigger breasts might see breasts roundness almost immediately, (did for me lol).

The CT therapy while noogling works, although I might suggest chilling the domes first. After using crushed ice inside the domes I pumped w/o ice, immediately the domes steamed up, exactly what I was hoping for, suggesting thermogenesis.

Another note: I see alternating an ice pack with warm compress for those not pumping could replace CT pumping. Use massage during and after to keep the blood flowing, the ice should raise heart rate for hormone release and activation of fat cells (BAT- brown adipose tissue).

Big Grin have fun with it.

(11-06-2016, 04:26 AM)Lotus Wrote:  I have some additional info relating to cold water/ice therapy talked about in previous post (below) specifically called Cold thermogenesis, CT for short. I think we step up CT by adding another option.........iced pumping, (noogling?, you get the picture). I also do a variation of taking ice packs directly over the breast while doing a cycle of isometrics (i.e, arms folded accross chest and flex each breast [separately] to the point of muscle fatigue).

The idea is to create new BAT (brown adipose tissue) by thermogenesis. Only in my opinion (lol) as we expend burned energy (via thermogenesis) fat is oxidized through mitochondria through HSP (heat shock proteins), some evidence has shown that when HSP's are increased it protects against oxidative stress, (this is postive news imo, similar to anti-inflammatories, only here, it's produced naturally in our cells). Now when I do this CT my heart rate immediately goes up, which activates this process too. Please seek medical advice before beginning any NBE regiment. I'm not a medical professional.

Combining cold thermogenesis with HIIT (high intensity interval training) aka depletion of energy in breast tissue could be of benefit. Add intermittent fasting to kick HGH (human growth hormone) in.

Quote:when you consume protein in levels higher than recommended above, you tend to activate the mTOR (mammalian target of rapamycin) pathway, which can help you get large muscles but may also increase your risk of cancer. There is also research suggesting that the "mTOR gene" is a significant regulator of the aging process, and suppressing this gene may be linked to longer life.
http://fitness.mercola.com/sites/fitness...rkout.aspx#!

Anyways lol. I'm getting ahead of myself lol, I'll have to break other related info over a couple of posts. Though some keys points are icing breasts while doing isometrics, try a cold version of pumping (crushed ice insides the domes), and do isometrics while pumping for fat burn.

The best part of this cold therapy (cold thermogenesis) is the low cost of ice water lol . And there's plenty of diy versions of ice packs.

Another version of thermogenesis happens when we sleep, natural melatonin plays a part of this. Adding melatonin at night (because we lose the natural production over time) with MSM looks very interesting, more info to follow: Big Grin

Proof of principle: the effects of chemical uncouplers:
The mitochondria of any organ of the body (except brown adipose tissue) are innately energy sparing through the ‘coupling’ of O2 consumption (i.e. thermogenesis) to ATP synthesis, only allowing the combustion of substrate when it is necessary, i.e. when ADP is available to be phos- phorylated (Fig. 2(A)). As heat is released when ATP is hydrolysed, thermogenesis can in principle occur either when ATP utilization is increased (e.g. in muscular work) or when protons are allowed to re-enter the mitochondria without ATP synthesis taking place, i.e. in ‘uncoupled’ mitochondria (Fig. 2(B and C)).

So I was looking into some research on cold water therapy and how it aids in recovery times in post HIIT workouts. Turns out that it also builds new brown fat cells, hmm, interesting huh?. Well.....it got me thinking on different ways to promote new cell growth for breast tissue. As we know fat cells produce aromatase enzyme, which converts testosterone into estrogen. We also know HIIT produces natural growth hormone and lactate, (initiates new muscle synthesis). It's quite possible (imo) using that cold water therapy and HIIT can trigger breast growth, the science says it's possible. Wink

Another example: From a clinical study I found that it stated that cAMP (2nd messengers) levels significantly increased lipolysis in adipose tissue, at 4°C, (39 degrees). Adipocytes increased the release of glycerol, indicating that the high rate of lipolysis occurred in these cells.

So how do this apply to NBE?, my opinion?..........hmm.......Rolleyes use ice packs on the boobs after massage/boobie exercises (you still do those grab n pull method boob exercises right?) I hope so cause that in-itself triggers breast growth. This method along with HIIT, cold water therapy is free, cept the cost of water. Rolleyes

Haha that is scary cool Boobi, this stuff drives you to new ideas and understanding for sure. For instance, this idea of what free energy does in metabolism makes thermogenesis so interesting, and specifically how we use adipose (fat tissue) to promote the body's own release of E2. The study below suggests we're not too far from pressing play without great difficulty (aka• aromatase to E2). Non-exercise activity thermogenesis.
Review article
Levine JA. Proc Nutr Soc. 2003.
Show full citation
Abstract
Non-exercise activity thermogenesis (NEAT) is the energy expended that is not from sleeping, eating or sports-like exercise. It ranges from the energy expended walking to work, typing, performing yard work, undertaking agricultural tasks and fidgeting. NEAT can be measured by one of two approaches. The first approach is to measure or estimate total NEAT. Here, total daily energy expenditure is measured and from it is subtracted BMR + thermic effect of food. The second is the factoral approach whereby the components of NEAT are quantified and total NEAT calculated by summing these components. The amount of NEAT that human subjects perform represents the product of the amount and types of physical activities and the thermogenic cost of each activity. The factors that affect the NEAT of a human subject are readily divisible into biological factors, such as weight, gender and body composition, and environmental factors, such as occupation or dwelling within a 'concrete jungle'. The combined impact of these factors explains the substantial variance in human NEAT. The variability in NEAT might be viewed as random but human data contradict this perception. It appears that changes in NEAT subtly accompany experimentally-induced changes in energy balance and are important in the physiology of weight change. NEAT and sedentariness may thus be important in obesity. It then becomes intriguing to dissect mechanistic studies that delineate how NEAT is regulated by neural, peripheral and humoral factors. NEAT may be a carefully-regulated 'tank' of physical activity that is crucial for weight control.

RIBOFLAVIN, A TESTOSTERONE 5a-REDUCTASE INHIBITOR

Riboflavin is a common product of several fungi and bacteria grown under proper conditions 2'3*.
It is used against riboflavin deficiency and hyperlipoproteinemia. It is reported here, for the first time, as a testosterone 5a-reductase inhibitor. (per the study).

Also, riboflavin is a co-factor of many oxido- reductases, while testosterone 5a-reductase utilizes the pyridine nucleotide, NADPH, to reduce testosterone. It is interesting that the co-factor for flavin enzymes, riboflavin, inhibits pyridine linked enzyme reaction.

Cool Wink...

I think 1-2 minutes hot application followed by 1-2 of ice (cold thermogenesis, CT). The quadrant (or clockwise ) technique works fast. Or try hot water first followed by a complete sweep of ice around the breast, though stopping at 1-2 per pole position. I'll tell what though, I think it jumped started a reaction that even made my hair stand on end lol. But definitely the boobs felt great. If you look at the web info on those products they state it helps relieves breast engorgement, mastitis and clogged ducts. (Perfect, a prolactin boost too lol).
